Overview

SYS6051 is an antibody-drug conjugate (ADC) developed by CSPC Megalith Biopharmaceutical, a subsidiary of CSPC Pharmaceutical Group, for the treatment of advanced solid tumors. The drug targets Protein Tyrosine Kinase 7 (PTK7), also known as Colon Carcinoma Kinase 4 (CCK4), a pseudo-kinase receptor that is overexpressed in various malignancies, including non-small cell lung cancer (NSCLC), esophageal cancer, and breast cancer, while maintaining limited expression in normal tissues. SYS6051 consists of a humanized anti-PTK7 monoclonal antibody conjugated to a potent cytotoxic payload. Upon binding to PTK7 on the tumor cell surface, the ADC is internalized through endocytosis, releasing the payload to induce cell cycle arrest and apoptosis. SYS6051 is currently in Phase I clinical development to evaluate its safety, pharmacokinetics, and preliminary efficacy in patients with refractory solid tumors.
