Overview

SYX-5219 is a novel, first-in-class oral small molecule therapy developed by Sitryx Therapeutics for the treatment of atopic dermatitis and potentially other inflammatory diseases. It acts as an activator of pyruvate kinase M2 (PKM2), a key regulator of immune cell metabolism. By shifting PKM2 into its anti-inflammatory state, SYX-5219 is designed to rebalance immune function, reduce inflammation, and restore skin barrier integrity in patients with atopic dermatitis. The drug modulates B and T lymphocyte activity at sites of inflammation, aiming to normalize immune cell function and drive sustained disease remission. Preclinical data indicate that SYX-5219 reduces inflammatory markers and enhances skin barrier repair. As of early 2025, it is undergoing Phase 1 clinical trials in healthy volunteers with plans to expand studies into adults with moderate to severe atopic dermatitis[1][3][4][7].
