Overview

SZ06531 is an extended-release (ER) small molecule loop diuretic under development by Sarfez Pharmaceuticals. It acts by inhibiting the sodium-potassium-chloride cotransporter (NKCC), primarily the NKCC2 isoform located in the thick ascending limb of the loop of Henle in the kidney. By blocking this transporter, the drug prevents the reabsorption of sodium, potassium, and chloride, leading to increased urine production (diuresis) and sodium excretion (natriuresis). This mechanism is utilized to manage fluid overload and congestion. SZ06531 is currently being evaluated in Phase 1 clinical trials for the treatment of heart failure, chronic kidney disease, and overactive bladder, with the extended-release formulation intended to provide a more controlled and sustained therapeutic effect compared to standard immediate-release diuretics.
