Overview

SZV558 is a **novel, potent, and selective monoamine oxidase B (MAO-B) inhibitor** classified as a (hetero)arylalkenyl propargylamine compound[1][3]. It exhibits **irreversible inhibition of the MAO-B enzyme**, protecting dopaminergic neurons through two mechanisms: inhibiting MAO-B enzymatic activity (thereby reducing dopamine breakdown and formation of toxic metabolites) and exerting neuroprotective effects independent of MAO-B inhibition, including protection against oxidative damage and toxic dopamine-quinone formation[1]. In preclinical models, SZV558 preserved striatal dopamine content after MPTP challenge, demonstrating superior efficacy compared to rasagiline at equivalent doses[1]. It has shown neuroprotective properties in vitro, with activity via both intraperitoneal and oral routes[1].
