T-AS1411 is an experimental, nucleolin-targeted drug delivery platform consisting of extracellular vesicles (EVs) or exosomes functionalized with the AS1411 aptamer. AS1411 is a synthetic G-quadruplex DNA aptamer that exhibits high affinity for nucleolin, a protein typically sequestered in the nucleolus but significantly overexpressed on the plasma membrane of various malignant cells, including breast cancer. By modifying the surface of EVs with AS1411, the platform achieves selective accumulation in tumor tissues. T-AS1411 is designed to encapsulate and protect therapeutic RNA molecules, such as let-7 microRNA or small interfering RNAs (siRNAs), from degradation in the systemic circulation, facilitating their delivery into the cytoplasm of target cells to modulate gene expression. Preclinical studies have demonstrated that T-AS1411-EVs can effectively target tumor tissues and inhibit tumor growth in breast cancer models with minimal non-specific toxicity or immunogenicity.
