**T-bet engineered iPSC-derived natural killer (NK) cells** are an investigational cell therapy derived from induced pluripotent stem cells (iPSCs), genetically modified to overexpress the transcription factor T-bet (TBX21). T-bet drives epigenetic activation of an NK cell-specific transcriptome, accelerating differentiation, enhancing maturation markers like CD16 and KIR, and improving functionality including IFN-γ production, cytotoxicity, and antibody-dependent cellular cytotoxicity (ADCC). This approach overcomes limitations of primary NK cells by enabling scalable, homogeneous "off-the-shelf" production for adoptive immunotherapy, primarily targeting cancers such as hepatocellular carcinoma (HCC), with preclinical evidence of superior anti-tumor activity in TGF-β-suppressed environments when combined with CARs targeting GPC3 or AFP[1][4].
