A specialized allogeneic hematopoietic stem cell transplantation (HSCT) procedure developed at the University of Wisconsin, Madison, for the treatment of relapsed lymphoma. This approach utilizes graft engineering to selectively deplete TCR-α/β+ T cells and CD19+ B cells from peripheral blood stem cell grafts obtained from haploidentical (half-matched) donors. By removing αβ T cells, which are the primary mediators of graft-versus-host disease (GVHD), while preserving γδ T cells and natural killer (NK) cells, the procedure aims to reduce the risk of GVHD and improve safety without requiring intensive pharmacological immunosuppression. The depletion of CD19+ B cells is further intended to prevent post-transplant Epstein-Barr virus (EBV)-associated lymphoproliferative disorders. This cell therapy modality is designed to provide a potent graft-versus-lymphoma effect while facilitating rapid hematopoietic engraftment.
