T-hIL12 is an oncolytic viral therapy based on a third-generation recombinant herpes simplex virus type 1 (HSV-1) engineered to express human interleukin-12 (IL-12). Developed by researchers at the University of Tokyo, the virus utilizes the G47Δ (G47delta) backbone, which features triple mutations—deletion of the γ34.5 and ICP47 genes, and inactivation of the ICP6 gene—to ensure selective replication within tumor cells while sparing healthy tissue. The local expression of IL-12 following intratumoral administration is designed to enhance the recruitment and activation of cytotoxic T lymphocytes and natural killer cells, thereby inducing a systemic anti-tumor immune response, often referred to as the abscopal effect. It is currently being investigated in clinical trials for patients with advanced malignant melanoma.
