T-MATE TCE anti-EGFR is a next-generation bispecific T-cell engager (TCE) developed by Amberstone Biosciences using its proprietary Tumor-Microenvironment Activated ThErapeutic (T-MATE) technology. This therapeutic is designed to selectively target the Epidermal Growth Factor Receptor (EGFR) in the acidic conditions characteristic of the tumor microenvironment (TME) while remaining inactive under normal physiological pH. By incorporating pH-dependent CD3-binding antibodies, the T-MATE platform aims to expand the therapeutic index of TCEs, minimizing on-target/off-tumor toxicities and cytokine release syndrome (CRS) typically associated with targeting widely expressed antigens like EGFR. Preclinical studies in humanized mouse models and non-human primates have demonstrated potent T-cell-dependent cytotoxicity and significant tumor regression across various solid tumor indications, including lung, colon, and breast cancers, with a favorable safety profile.
