Overview

T-MATE TCE anti-TROP2 is a preclinical-stage bispecific T-cell engager (TCE) developed by Amberstone Biosciences using its proprietary Tumor-Microenvironment Activated ThErapeutic (T-MATE) platform. The drug is designed to target Trophoblast cell-surface antigen 2 (TROP2), a protein frequently overexpressed in various solid tumors, while minimizing systemic toxicity. It incorporates a pH-dependent CD3-binding domain that remains inactive at physiological pH but undergoes a conformational change to bind CD3 in the acidic conditions characteristic of the tumor microenvironment. This selective activation is intended to direct T-cell cytotoxicity specifically to tumor tissues, thereby reducing the risk of cytokine release syndrome (CRS) and on-target/off-tumor toxicities in healthy tissues where TROP2 is also expressed.
