T-MATE TCE FOLR1 is a bispecific T-cell engager (TCE) developed by Amberstone Biosciences using its proprietary Tumor-Microenvironment Activated ThErapeutic (T-MATE) platform. The drug is designed to target Folate Receptor 1 (FOLR1) on tumor cells and the CD3 complex on T cells, facilitating T-cell-mediated cytotoxicity against cancer cells. A distinguishing feature of the T-MATE platform is its pH-dependent activation; the antibody is engineered to bind its targets with high affinity in the acidic conditions characteristic of the tumor microenvironment (TME) while exhibiting minimal activity at normal physiological pH. This selectivity is intended to expand the therapeutic index by reducing on-target/off-tumor toxicities in healthy tissues and mitigating systemic cytokine release syndrome (CRS). Preclinical data presented at AACR 2025 demonstrated potent activity and significant tumor regression in xenograft models across multiple solid tumor indications, including ovarian, endometrial, and non-small cell lung cancers.
