T-Plex is an autologous, multiplexed T cell receptor-engineered T cell (TCR-T) therapy developed by TScan Therapeutics for the treatment of various solid tumors. The therapy utilizes a customized combination of two to three different TCR-T cell products selected from TScan's ImmunoBank, a repository of therapeutic TCRs tailored to a patient's specific HLA type and tumor antigen expression. T-Plex targets multiple cancer-associated antigens, including MAGE-A1, MAGE-A4, MAGE-C2, PRAME, and HPV16 E7, presented by various HLA class I molecules such as HLA-A*02:01, HLA-A*01:01, HLA-C*07:02, and HLA-B*07:02. Each T cell component is engineered using a non-viral transposon-based vector to express a therapeutic TCR along with CD8α/β co-receptors to enhance potency in CD4+ T cells, a CD34 epitope tag for selection and tracking, a dominant-negative TGFβ receptor II (dnTGFβRII) to resist immunosuppression in the tumor microenvironment, and a mutated dihydrofolate reductase (DHFR) for potential selection. This multiplexed approach is designed to overcome tumor heterogeneity and HLA loss of heterozygosity, which are common resistance mechanisms in solid tumor immunotherapy.
