Drug intelligence / Profile preview
T-sema3A is a recombinant, engineered, truncated form of the endogenous immunoregulatory protein semaphorin-3A (sema3A). Originally discovered and designed at the Technion - Israel Institute of Technology, it was developed by the spinout company Symbol Therapeutics. T-sema3A is engineered to selectively bind and signal through the immune-regulatory CD72 receptor on B and T lymphocytes while lacking the C-terminal domain required to bind neuropilin-1 (NRP-1), thereby avoiding the unwanted endothelial and neuronal effects associated with wild-type sema3A. Upon binding CD72, T-sema3A down-regulates STAT-4 and HDAC-1 phosphorylation and up-regulates PKC-theta and p38-MAPK phosphorylation. This signaling cascade promotes the expression of anti-inflammatory regulatory cytokines (such as IL-10 and TGF-beta) and suppresses pro-inflammatory cytokines (such as IL-6, IFN-gamma, and IL-17A), aiming to restore self-tolerance and immune homeostasis in autoimmune and inflammatory diseases. Symbol Therapeutics ceased operations and entered liquidation in November 2025, leading to the discontinuation of the program.
