Overview

**T-siHER2-NP** is a preclinical, trastuzumab-targeted mesoporous silica nanoparticle formulation for drug-resistant **HER2-positive breast cancer**. The construct uses trastuzumab to bind HER2-positive tumor cells and promote cellular uptake while delivering a small interfering RNA directed against **ERBB2/HER2** messenger RNA, resulting in HER2 protein knockdown and suppression of HER2-dependent signaling. In preclinical HER2-positive breast cancer models, the formulation showed activity against tumors resistant to trastuzumab and taxanes, including HCC1954 and BT474-TR models. It was developed from nanoparticle work at Oregon Health & Science University and is being advanced by PDX Pharmaceuticals.
