Overview

T-VISA-PEA-15 is a targeted gene therapy construct developed for advanced breast cancer, utilizing a breast cancer-specific vector termed T-VISA—which incorporates the human telomerase reverse transcriptase (hTERT) promoter and a transgene amplifier system (VISA: VP16-GAL4-WPRE). This system drives tumor-selective expression of PEA-15 (phosphoprotein enriched in astrocytes), a protein containing a death effector domain that sequesters extracellular signal-regulated kinase (ERK) in the cytoplasm, thereby inhibiting cell proliferation and inducing apoptosis. T-VISA-PEA-15 is delivered using liposome nanoparticles, showing high specificity for breast cancer cells, robust induction of cancer cell death in vitro and in vivo, and minimal toxicity to normal cells. Preclinical models demonstrate strong suppression of tumor growth and prolonged survival in mice, without severe adverse effects. The mechanism centers on targeted induction of apoptosis, reduction of ERK signaling, and tumor-specific gene amplification, making T-VISA-PEA-15 a promising clinical candidate for breast cancer gene therapy[1][3][5][7][10].
