T001

T001 is a preclinical small-molecule senoblocker developed by Dorian Therapeutics (acquired by Altos Labs) to optimize T cell therapies for cancer treatment. It functions as an inhibitor of the deubiquitinating enzyme USP16, an epigenetic regulator that plays a key role in cellular senescence. By modulating USP16 during the ex vivo expansion of T cells, T001 prevents the premature aging and exhaustion of the cells, thereby enhancing their fitness, persistence, and therapeutic efficacy upon re-infusion into the patient. This approach addresses a major limitation in adoptive cell transfer where T cells often lose functionality during the manufacturing process. The technology originated from research at Stanford University and was developed in collaboration with Ubiquigent.
