Overview

**T1367** is an autologous T-cell receptor (TCR)-based cell therapy engineered to target the MAGE-A1 cancer/testis antigen, an intracellular protein presented by HLA-A*02:01 on multiple myeloma (MM) cells. Derived from T-knife Therapeutics' proprietary humanized TCR (HuTCR) mouse platform, TCR-T1367 T cells are manufactured by transducing patient-derived T cells with a retrovirus encoding the TCR specific for the MAGE-A1₂₇₈-₂₈₆ epitope, enabling recognition and lysis of MAGE-A1-expressing tumor cells with high antigen sensitivity compared to CAR-T therapies. Developed for relapsed/refractory MM patients with ≥30% MAGE-A1-positive cells, particularly those with extramedullary disease or post-BCMA-targeted therapy relapse, a phase I trial (EudraCT 2017-001208-30) at Charité-Universitätsmedizin Berlin treated two patients at 10⁵ cells/kg, showing feasibility with no severe treatment-related adverse events but limited efficacy due to low dosing and early trial closure; manufacturing occurred at the GMP facility of the Experimental and Clinical Research Center (ECRC).
