Drug intelligence / Profile preview
T1D-tolLNP (also known as INT-101) is a preclinical-stage, antigen-specific immune tolerance therapy (ASIT) developed by Integrated Nanotherapeutics in collaboration with the University of British Columbia. Designed as a tolerogenic mRNA vaccine for type 1 diabetes (T1D), it utilizes a proprietary multi-cargo lipid nanoparticle (LNP) platform (ScaffoLD lipid technology) to co-deliver mRNA encoding multiple beta-cell autoantigens (such as proinsulin) along with tolerogenic small-molecule immunomodulators. By delivering these components simultaneously to antigen-presenting cells, T1D-tolLNP aims to retrain the immune system, promoting the expansion of antigen-specific regulatory T cells (Tregs) and blunting the autoimmune destruction of insulin-producing pancreatic beta cells. Preclinical studies in non-obese diabetic (NOD) mice have demonstrated that T1D-tolLNP can prevent the onset of autoimmune diabetes and reverse new-onset disease, restoring sustained euglycemia without the need for chronic immunosuppression.
