T3D-959 is an orally administered small molecule that acts as a dual agonist of peroxisome proliferator-activated receptor delta (PPARδ) and gamma (PPARγ), with a 15-fold higher potency for PPARδ. It is the first PPARδ activator developed specifically for Alzheimer's disease and also shows activity at PPARγ, which may provide additive or synergistic effects on brain glucose and lipid metabolism. Originally developed for diabetes and dyslipidemia under the name DB959, it was later acquired by T3D Therapeutics from DARA BioSciences in 2013 and repurposed for neurodegenerative diseases. The drug has demonstrated potential to improve dysfunctional glucose and lipid metabolism in the brain, reduce neuroinflammation, mitigate cognitive decline related to cerebral metabolic dysfunction, and impact biomarkers associated with Alzheimer’s disease. Clinical trials have shown safety as well as efficacy signals in slowing clinical decline in Alzheimer’s patients[1][2][4][5][8].
