T5244 is a small molecule inhibitor of the c-Fos/AP-1 transcription factor complex. It is being investigated for its potential to overcome temozolomide (TMZ) resistance in glioblastoma (GBM), particularly in patients with unmethylated MGMT. The drug works by inhibiting the activation of Activator Protein-1 (AP-1), which in turn prevents the upregulation of cancer stem cell (CSC) markers such as CD44 and ALDH1A1. These markers are typically induced by the metastasis-enhancing protein KITENIN and are associated with tumor progression and poor clinical outcomes. By blocking this pathway, T5244 aims to reduce tumor recurrence and improve survival in aggressive brain cancers.
