T8Ab is a cross-species neutralizing monoclonal antibody that targets Anthrax Toxin Receptor 1 (ANTXR1), also known as Tumor Endothelial Marker 8 (TEM8). Originally developed for cancer therapy due to the overexpression of ANTXR1 in tumor vasculature and certain cancer cells, T8Ab is being investigated for its potent antifibrotic and cardioprotective properties. In the context of heart disease, ANTXR1 is identified as a master regulator of fibrosis, overexpressed in diseased cardiac fibroblasts. T8Ab functions by blocking ANTXR1, which leads to the suppression of fibrosis-related genes (such as Ccn2, Ccn4, and Pdgfa) and the reduction of maladaptive TGF-β-driven extracellular matrix (ECM) remodeling. Preclinical studies have demonstrated its efficacy in improving cardiac function and reducing scar size in models of myocardial infarction, hypertension-induced heart disease, and heart failure with preserved ejection fraction (HFpEF).
