Overview

**TA-CD** is an investigational active immunotherapy for cocaine dependence. It is a conjugate vaccine comprising the cocaine hapten **succinylnorcocaine** coupled to recombinant cholera toxin B subunit and adsorbed to aluminum hydroxide adjuvant. Intramuscular vaccination elicits cocaine-specific circulating antibodies that bind cocaine in blood, reducing the free drug available to cross the blood-brain barrier and thereby attenuating its reinforcing and subjective effects. The program originated as IPC-1010 at ImmuLogic Pharmaceutical, was subsequently developed by Xenova, and later by Celtic Pharma. Clinical studies found highly variable antibody responses; participants achieving higher anti-cocaine antibody titers showed reduced subjective effects of smoked cocaine and improved cocaine-free urine outcomes in a subgroup.
