TA8Amino is a novel multi-target small molecule hybrid compound derived from donepezil and tacrine, designed for the treatment of Alzheimer's disease. It acts as a dual inhibitor of acetylcholinesterase (AChE) and Rho-associated protein kinase 2 (ROCK2). By inhibiting ROCK2, TA8Amino promotes neurogenesis through the activation of the PTEN/AKT signaling pathway, as demonstrated in SH-SY5Y neuroblastoma cells. Unlike its parent compounds, TA8Amino significantly reduces ROCK2 protein expression and increases neurite length and neurodifferentiation markers such as β-III-tubulin and MAP2.
