Overview

TA99-CAR T cells are a type of chimeric antigen receptor (CAR) T cell therapy specifically engineered to target **tyrosinase-related protein 1 (TRP1, also known as TYRP1)**, a protein expressed on melanocytes and melanoma cells. The CAR comprises a single-chain variable fragment (scFv) from the TA99 antibody, linked to CD28 transmembrane and costimulatory domains, and a CD3ζ signaling domain. This engineered T cell therapy is designed to recognize and kill TRP1-expressing tumor cells. Preclinical studies in mice have shown that TA99-CAR T cells are capable of cytotoxicity against melanoma cells in vitro, but as a single agent, have limited in vivo efficacy against established solid tumors in immunocompetent models. Research is ongoing to enhance their efficacy, including arming TA99-CAR T cells with additional cytokines (e.g., IL-2 variants, IL-33) to overcome the immunosuppressive tumor microenvironment and improve anti-tumor immunity. The main indication is for **melanoma**, particularly aggressive and immune-resistant forms[1][2].
