TAB004 + gemcitabine + cisplatin + MSN is an experimental targeted nanotherapeutic system designed for the treatment of cancers expressing the tumor-associated form of Mucin 1 (tMUC1), such as pancreatic ductal adenocarcinoma (PDA). The system utilizes mesoporous silica nanoparticles (MSN) as a carrier to co-deliver the cytotoxic agents gemcitabine and cisplatin. The surface of the MSN is functionalized with TAB004, a monoclonal antibody that specifically binds to tMUC1, which is overexpressed and hypoglycosylated in over 80% of human pancreatic cancers but not in normal tissues. This targeting mechanism aims to enhance the accumulation of the chemotherapy drugs within the tumor microenvironment while reducing systemic toxicity. Gemcitabine acts as a nucleoside analog that inhibits DNA synthesis by targeting ribonucleoside reductase, while cisplatin is a platinum-based agent that causes DNA cross-linking, leading to apoptosis.
