Tabituximab barzuxetan is a radiolabeled antibody-drug conjugate designed for targeted cancer therapy, primarily in synovial sarcoma and other soft tissue sarcomas. It consists of a humanized monoclonal antibody (OTSA101) that targets the Frizzled Homolog 10 receptor (FZD10), which is highly expressed on certain tumor cells. The antibody is conjugated to a chelator (DTPA), allowing it to be labeled with the radioactive isotope yttrium-90. Upon administration, the drug binds specifically to FZD10-positive tumor cells and delivers cytotoxic beta radiation directly to these cells, resulting in targeted cell death while sparing surrounding healthy tissue. Tabituximab barzuxetan has been granted orphan drug status by both the European Commission and US FDA and is currently being evaluated in early-phase clinical trials for soft tissue sarcoma[1][3][5].
