Overview

TAC-001 is an investigational, systemically delivered antibody-drug conjugate developed for the treatment of solid tumors. It is a Toll-like Receptor Agonist Antibody Conjugate (TRAAC) comprised of a potent toll-like receptor 9 (TLR9) agonist oligonucleotide (T-CpG) conjugated to an antibody targeting CD22, a receptor restricted to B cells, including tumor-infiltrating B cells. By binding to CD22 and being internalized by B cells, TAC-001 delivers the TLR9 agonist directly into these immune cells, leading to their activation and triggering both innate and adaptive anti-tumor immune responses. Preclinical studies have shown that this mechanism results in increased B cell infiltration in tumors, enhanced T cell effector function, modulation of suppressive myeloid cells within the tumor microenvironment, and robust anti-tumor activity even in checkpoint inhibitor-resistant models. Early clinical data from Phase 1/2 trials indicate that TAC-001 is well tolerated with preliminary evidence of durable disease stabilization and partial responses in patients with advanced or metastatic solid tumors[1][2][3][4][5].
