Overview

TAC-401 (also known as TP-0401) is an investigational potassium-competitive acid blocker (P-CAB) developed by Taiho Pharmaceutical for the treatment of acid-related disorders and the eradication of *Helicobacter pylori* infection. As a P-CAB, TAC-401 inhibits the gastric H+,K+-ATPase (the proton pump) in a potassium-competitive and reversible manner. Compared to traditional proton pump inhibitors (PPIs), P-CABs offer a faster onset of action, more consistent and potent acid suppression, and do not require activation by gastric acid. In the context of *H. pylori* eradication, TAC-401 is used to maintain a high intragastric pH, which enhances the stability and antimicrobial activity of co-administered antibiotics. It is currently undergoing Phase 2 clinical development in Japan.
