Overview

TAC-T cells are a novel type of engineered T-cell therapy designed for cancer treatment, particularly solid tumors. This therapy utilizes a T-cell antigen coupler (TAC) receptor, which differs from traditional CAR-T cells by co-opting the endogenous T-cell receptor (TCR) signaling pathway. This mechanism aims to achieve robust anti-tumor activity while potentially offering an improved safety profile by mimicking natural T-cell activation and reducing off-target toxicities and cytokine release syndrome. The specific TAC-T cells in question are engineered to target Nectin4/NKG2DL and FAP, and are further modified to secrete cytokines such as Interleukin-7 (IL7), Chemokine (C-C motif) ligand 21 (CCL21), and Interleukin-12 (IL12) to enhance their anti-tumor efficacy and overcome the immunosuppressive tumor microenvironment.
