Overview

TAC01-CLDN18.2 is a novel autologous cell therapy consisting of genetically engineered T cells that express a proprietary chimeric receptor called the T-cell Antigen Coupler (TAC), which specifically recognizes Claudin 18.2 (CLDN18.2), a tight junction protein aberrantly expressed in several solid tumors including gastric, gastroesophageal, esophageal adenocarcinoma, non–small cell lung cancer, cholangiocarcinoma, pancreatic ductal adenocarcinoma, ovarian mucinous cancer, and colorectal cancer[1][4][7]. Upon engagement with CLDN18.2 on tumor cells, the engineered T-cells are activated via their endogenous T cell receptor to mediate targeted anti-tumor responses[1][5][7]. The therapy is being developed as an immunotherapy for patients with unresectable or metastatic CLDN18.2-positive solid tumors and is currently under investigation in phase 1/2 clinical trials[3][4].
