TAC01-HER2 + pembrolizumab is an investigational combination therapy for HER2-positive solid tumors, particularly gastric and gastroesophageal junction (GEJ) adenocarcinoma. TAC01-HER2 is an autologous cell therapy consisting of genetically engineered T cells expressing a T-cell Antigen Coupler (TAC) that specifically recognizes human epidermal growth factor receptor 2 (HER2). Upon engagement with HER2, the TAC activates the T cells via their endogenous T cell receptor, directing them to attack HER2-expressing tumor cells. Pembrolizumab is a monoclonal antibody immune checkpoint inhibitor targeting programmed death-1 (PD-1), thereby blocking inhibitory PD-L1/PD-1 signaling and enhancing anti-tumor immune responses. The combination aims to leverage both targeted cellular cytotoxicity against HER2-positive cancer cells and release of immune inhibition by PD-1 blockade. This regimen is being evaluated in clinical trials for relapsed or refractory HER2-positive solid tumors[3][4][5][6].
