Tacalcitol is a synthetic vitamin D3 analog used topically for the treatment of mild to moderate plaque psoriasis and other skin conditions characterized by excessive keratinocyte proliferation, such as chronic chapped lips and severe dry skin. It works primarily by binding to vitamin D receptors on keratinocytes in the epidermis, inhibiting their hyperproliferation and inducing differentiation. Tacalcitol also modulates inflammatory responses in the skin by downregulating cutaneous lymphocyte-associated antigen (CLA) expression on T cells, thereby reducing T-cell infiltration and inflammation. Developed to maintain potent cell-regulating properties of calcitriol with fewer calcium-related side effects, tacalcitol is available as ointment or lotion formulations for topical use[1][2][4][5][6][7].
