TACE + acoradine is a combination therapy regimen primarily investigated for the treatment of hepatocellular carcinoma (HCC). It combines Transarterial Chemoembolization (TACE), a locoregional interventional procedure, with acoradine (icaritin), an orally active small molecule. TACE involves the targeted delivery of chemotherapeutic agents (such as doxorubicin or cisplatin) into the tumor's blood supply via the hepatic artery, followed by the administration of embolic agents to induce ischemic necrosis. Acoradine is a prenylated flavonoid derived from the *Epimedium* genus that acts as a first-in-class inhibitor of the estrogen receptor-alpha 36 (ER-α36) variant and modulates the IL-6/JAK/STAT3 signaling pathway to exert immunomodulatory and anti-tumor effects. The combination is designed to leverage the immediate debulking effect of TACE with the sustained systemic activity of acoradine to improve overall survival and reduce recurrence in patients with intermediate or advanced HCC.
