TACRO-Skin is an investigational topical 0.1% tacrolimus microemulsion solution specifically developed by Bay Pharma GmbH for the treatment of scalp psoriasis. The formulation utilizes a kerato-emulsifying microemulsion technology designed to enhance the cutaneous bioavailability and penetration of tacrolimus through the thick hyperkeratotic plaques characteristic of scalp psoriasis. Tacrolimus, the active ingredient, is a macrolide calcineurin inhibitor that suppresses T-lymphocyte activation and the subsequent production of pro-inflammatory cytokines (such as IL-2) by inhibiting the phosphatase activity of calcineurin. In a multicenter Phase 3 clinical trial, TACRO-Skin demonstrated statistically significant efficacy over vehicle placebo in achieving clear or almost clear scalp conditions (IGA 0 or 1) after 8 weeks of treatment.
