tacrolimus (Sandoz)

Dailiport is a once-daily, prolonged-release oral formulation of tacrolimus, a calcineurin inhibitor immunosuppressant. Developed by Sandoz as a branded generic of Advagraf, Dailiport is indicated for the prophylaxis of transplant rejection in adult kidney or liver allograft recipients, as well as for the treatment of allograft rejection resistant to other immunosuppressive regimens. By binding to the intracellular immunophilin FKBP12, tacrolimus forms a complex that inhibits calcineurin, a calcium/calmodulin-dependent phosphatase. This inhibition prevents the dephosphorylation and nuclear translocation of nuclear factor of activated T-cells (NFAT), thereby blocking T-cell activation and the transcription of key cytokines like interleukin-2 (IL-2).
