TACTIK trial strategy

The TACTIK trial (GETUG-AFU 28) is a Phase 2 clinical study sponsored by UNICANCER designed to evaluate a personalized treatment approach for patients with metastatic castration-resistant prostate cancer (mCRPC). The strategy utilizes circulating tumor cell (CTC) counts as a biomarker for early docetaxel resistance. Patients with ≥5 CTCs/7.5 mL after two cycles of docetaxel are randomized to either continue docetaxel (75 mg/m²) or switch to cabazitaxel (20 mg/m²). Both docetaxel and cabazitaxel are taxane-derived small molecules that function by binding to and stabilizing microtubules, thereby inhibiting mitotic cell division and inducing apoptosis in cancer cells. This trial represents a treatment strategy rather than the development of a novel therapeutic agent.
