TACTIVA CD4 is a preclinical-stage adoptive cell therapy developed by Tactiva Therapeutics for the treatment of solid tumors expressing the NY-ESO-1 cancer-testis antigen. The therapy utilizes hematopoietic stem cell (HSC)-derived CD4 T cells engineered to express a high-affinity T-cell receptor (TCR) specific for NY-ESO-1. Unlike traditional TCR-T therapies that focus primarily on mature CD8+ cytotoxic T cells, this approach leverages the unique properties of HSC-derived CD4+ cells to provide a long-term, self-renewing source of tumor-specific T cells. These cells are designed to provide sustained "cleanup" of remaining tumor cells and immune protection against cancer recurrence, complementing CD8-mediated tumor killing to achieve durable remissions in solid malignancies.
