TadCBEd mRNA is a messenger RNA-based gene editing therapeutic candidate that encodes a high-fidelity, TadA-derived cytosine base editor (CBE). Developed to overcome the limitations of traditional APOBEC-based cytosine base editors—such as significant DNA and RNA off-target activity—TadCBEd utilizes an engineered version of the E. coli adenosine deaminase TadA that has been evolved to deaminate cytosine instead of adenine. This specific variant, TadCBEd, incorporates mutations designed to minimize off-target effects while maintaining high editing efficiency for C-to-T transitions. Delivered as an mRNA, the editor is expressed transiently within the target cells, facilitating precise genomic modifications without the requirement for double-strand breaks or the long-term presence of the editing machinery. This technology is primarily being explored for the treatment of various genetic disorders caused by point mutations that can be corrected via cytosine-to-thymine conversion.
