Drug intelligence / Profile preview
Tafamidis meglumine (also known by the Chinese generic name chlorbenzoxamic acid meglumine) is a small molecule kinetic stabilizer of transthyretin (TTR). It functions by binding with high affinity and selectivity to the two thyroxine-binding sites on the TTR tetramer. This binding prevents the dissociation of the tetramer into monomers, which is the rate-limiting step in the amyloidogenic process. By stabilizing the tetrameric form of TTR, the drug inhibits the formation of amyloid fibrils that lead to tissue damage in transthyretin amyloidosis. Qilu Pharmaceutical has developed a generic version of tafamidis meglumine in China, which is currently being evaluated in a Phase IV clinical trial (CTR20251216) to assess its long-term safety and effectiveness in adult patients with stage 1 transthyretin amyloid polyneuropathy (ATTR-PN).
