Tafasitamab is a humanized, Fc-modified monoclonal antibody that targets the CD19 antigen expressed on the surface of pre-B and mature B lymphocytes, as well as several B-cell malignancies including diffuse large B-cell lymphoma (DLBCL). Upon binding to CD19, tafasitamab mediates B-cell lysis through direct induction of apoptosis and immune effector mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). The Fc region of tafasitamab is engineered for enhanced affinity to Fcγ receptors, increasing its potency in engaging immune effector cells like natural killer cells and γδ T cells. Tafasitamab is approved in combination with lenalidomide for adults with relapsed or refractory DLBCL who are not eligible for autologous stem cell transplant[2][4][5][6][8].
