Tafasitamab + lenalidomide + gemcitabine + oxaliplatin is a four-drug combination regimen under investigation for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL). - **Tafasitamab** is a humanized monoclonal antibody targeting CD19 on B cells, engineered to enhance antibody-dependent cellular cytotoxicity and phagocytosis, as well as to directly induce cell death via B-cell receptor signaling. - **Lenalidomide** is an immunomodulatory agent that stimulates T cell and natural killer cell function and induces direct tumor cell death, modifying the immune microenvironment. - **Gemcitabine** is a nucleoside analog acting as an antimetabolite interfering with DNA synthesis, inducing apoptosis in dividing cells. - **Oxaliplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA replication and transcription leading to cell death. This four-drug combination is currently being studied in a randomized, open-label, phase 3 clinical trial for patients with relapsed or refractory DLBCL, not eligible for autologous stem cell transplant, comparing it directly to rituximab plus gemcitabine and oxaliplatin[5].
