TAG-72 CAR T is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed to target Tumor-Associated Glycoprotein 72 (TAG-72), a pan-adenocarcinoma oncofetal antigen highly expressed on the surface of various solid tumors, including ovarian, colorectal, breast, and pancreatic cancers. Developed primarily by City of Hope, the therapy involves genetically engineering a patient's own T cells to express a CAR construct. This construct typically features a humanized single-chain variable fragment (scFv) derived from the CC49 or B72.3 antibodies, which provides high-affinity binding to TAG-72, linked to intracellular signaling domains such as CD3-zeta and co-stimulatory domains like 4-1BB (CD137) or CD28. The therapy is currently being evaluated in Phase 1 clinical trials for advanced epithelial ovarian cancer and has been studied in metastatic colorectal cancer, aiming to overcome the challenges of the immunosuppressive tumor microenvironment in solid malignancies.
