TAG-T cells targeting KRAS G12D are an experimental allogeneic cell therapy utilizing gamma-delta (γδ) T cells engineered with a T-cell receptor mimic (TCRm) based activating receptor (TAG). This platform specifically targets the intracellular KRAS G12D mutation when presented as a peptide-MHC complex by the HLA-A*11 allele. The TAG receptor is designed as a double-chain, TCR-based construct where a KRAS G12D/HLA-A*11-specific single-chain variable fragment (scFv), identified via phage display, is fused to human TCR-Cγ and TCR-Cδ chains. To enhance therapeutic potency and persistence within the immunosuppressive tumor microenvironment, these cells are further engineered with co-stimulatory domains and a chimeric switching receptor (CSR). This approach aims to provide a potent, off-the-shelf treatment option for patients with KRAS G12D-positive cancers, particularly those carrying the HLA-A*11 genotype.
