The TAG vaccine is an autologous, gene-modified tumor cell-based immunotherapy developed by the Mary Crowley Medical Research Center in collaboration with Gradalis. It is a 'triad vaccine' designed to overcome immune evasion in advanced cancers. The vaccine consists of the patient's own tumor cells transfected with a dual-expression plasmid containing a TGFβ2 antisense gene and a GM-CSF gene. GM-CSF serves to stimulate the recruitment and maturation of dendritic cells at the injection site, enhancing tumor antigen presentation. Simultaneously, the TGFβ2 antisense component knocks down the expression of TGFβ2, a potent immunosuppressive cytokine often secreted by tumors to inhibit T-cell activity. This dual approach aims to both prime the immune system and remove local inhibitory signals, facilitating a more robust anti-tumor response in patients with advanced metastatic solid tumors.
