TAG72CAR T cells are chimeric antigen receptor (CAR) T cells designed to target **tumor-associated glycoprotein 72 (TAG-72)**, a pan-adenocarcinoma antigen highly expressed in cancers such as ovarian, colorectal, breast, prostate, and stomach. The CAR construct utilizes a single-chain variable fragment (scFv) specific for TAG-72, often derived from the CC49 monoclonal antibody, and couples this to intracellular signaling domains including CD28 or 4-1BB and CD3ζ. Second-generation constructs enhance tumor cell recognition and killing by incorporating improved affinity scFv and optimized costimulatory regions[1][2]. These CAR-T cells are investigated primarily for solid tumors expressing TAG-72, with particular focus on ovarian and colorectal cancers.
