Tagileridine (SHR8554) is a novel, small-molecule mu-opioid receptor (MOR) agonist developed by Jiangsu Hengrui Pharmaceuticals for the management of acute pain. It is designed as a biased ligand that preferentially activates the G-protein signaling pathway over the β-arrestin-2 recruitment pathway. This biased mechanism of action is intended to provide potent analgesic efficacy while potentially reducing the incidence of typical opioid-related adverse effects, such as respiratory depression, nausea, and gastrointestinal dysfunction. Tagileridine is primarily being investigated for the treatment of postoperative pain and the prevention of remifentanil-induced hyperalgesia.
