Tagileridine fumarate (SHR8554) is a novel, potent, and selective G-protein biased agonist of the mu-opioid receptor (MOR). Developed by Jiangsu Hengrui Pharmaceuticals, it is designed to provide effective analgesia for moderate-to-severe acute pain while minimizing the recruitment of beta-arrestin 2, a pathway associated with typical opioid-related adverse events (ORAEs) such as respiratory depression and gastrointestinal dysfunction. By selectively activating the G-protein signaling pathway, tagileridine aims to offer a wider therapeutic window compared to traditional opioids like morphine or sufentanil. It was approved by the China National Medical Products Administration (NMPA) in May 2023 for the treatment of moderate-to-severe acute pain, particularly in the postoperative setting.
