Tagilidine (SHR8554) is a novel, potent, and selective biased mu-opioid receptor (MOR) agonist developed by Jiangsu Hengrui Pharmaceuticals. It is designed to selectively activate the G-protein signaling pathway while minimizing the recruitment of β-arrestin-2, a mechanism intended to provide effective analgesia for moderate to severe postoperative pain while reducing opioid-related adverse effects such as respiratory depression and gastrointestinal dysfunction. Tagilidine fumarate was approved by the China National Medical Products Administration (NMPA) in May 2023 for the treatment of moderate to severe postoperative pain. It is typically administered via intravenous injection or patient-controlled intravenous analgesia (PCIA).
