TAK-003 (marketed as Qdenga) is a live-attenuated tetravalent vaccine developed by Takeda for the prevention of dengue fever caused by any of the four dengue virus serotypes (DENV-1, DENV-2, DENV-3, and DENV-4). The vaccine is constructed using a live-attenuated dengue serotype 2 virus (strain PDK-53) as the genetic backbone. To provide protection against the other three serotypes, the pre-membrane (prM) and envelope (E) genes of the DENV-2 backbone are replaced with the corresponding genes from DENV-1, DENV-3, and DENV-4, resulting in a chimeric formulation. TAK-003 triggers both neutralizing antibody production and cross-reactive T-cell-mediated immune responses. It is administered subcutaneously in a two-dose schedule, three months apart, and has demonstrated sustained efficacy and safety in Phase 3 trials, particularly in preventing hospitalized dengue cases.
