TAK-044 is a **peptide-based, combined endothelin-A and endothelin-B receptor antagonist** developed by Takeda. It was designed to block the effects of endothelin-1 (ET-1), a potent vasoconstrictor implicated in various cardiovascular and renal diseases. The compound demonstrated systemic vasodilation, reduced blood pressure, and improved cardiac and renal hemodynamics in preclinical and clinical studies. TAK-044 showed cardioprotective effects in myocardial ischemia/reperfusion injury models, preserving ATP levels and reducing tissue damage. Despite promising early results, development was discontinued in phase II clinical trials for multiple indications including myocardial infarction, acute renal failure, acute hepatic malfunction, and subarachnoid hemorrhage. The compound had a short half-life but produced sustained endothelin receptor blockade lasting up to 12 hours after administration.
